PanCAN Funds Sylvester Study to Validate Blood Test for Pancreatic Cancer

The Pancreatic Cancer Action Network (PanCAN) has awarded a $225,000 grant to researchers at Sylvester Comprehensive Cancer Center, part of the University of Miami Miller School of Medicine. This two-year award will support a study aimed at validating circulating tumor DNA (ctDNA) blood tests as a reliable method for determining the molecular makeup of metastatic pancreatic cancer. Currently, patients with pancreatic cancer primarily rely on more invasive and time-consuming tissue biopsies for molecular profiling. The Sylvester team, led by principal investigator Peter Hosein, M.D., and co-principal investigator Jashodeep Datta, M.D., will utilize patient samples from PanCAN’s Precision Promise adaptive clinical trial. They will compare ctDNA results from blood samples with those obtained from standard tissue testing from the same patients. A study conducted by researchers at Western University's Schulich School of Medicine & Dentistry and London Health Sciences Centre Research Institute (LHSCRI) has revealed that advanced genetic testing can significantly broaden treatment options for cancer patients. The prospective study, named Precision Oncology at Western University (POWER) and published in The Journal of Molecular Diagnostics, utilized next-generation sequencing to analyze hundreds of cancer-related genes simultaneously. This approach uncovered genetic changes that standard-of-care testing often misses. The findings indicate that nearly one in three patients became eligible for therapies not typically used for their specific cancer type, based on the unique DNA of their tumor. This expanded testing also provided access to clinical trials, off-label therapies, and compassionate access programs that were previously unavailable to these patients.

Taiho Oncology and Cullinan Therapeutics are moving forward with plans to file for FDA approval of their oral drug candidate, zipalertinib, for non-small cell lung cancer (NSCLC), despite three patient deaths reported during a late-stage study. The Phase 3 REZILIENT3 trial demonstrated that zipalertinib, when combined with chemotherapy, significantly improved progression-free survival (PFS) in patients with first-line advanced non-squamous NSCLC harboring exon 20 mutations to the EGFR gene. Patients receiving the zipalertinib regimen experienced a 50% reduction in the risk of death or disease progression, with a median PFS of 14.5 months compared to 8.5 months in the control group. While adverse events of grade 3 or higher were more frequent in the zipalertinib arm (87% vs. 54.4%), analysts believe the robust efficacy and potential mitigation strategies will outweigh the safety concerns.
