PinkyCaMP Sensor Enhances Neural Bioanalysis with Improved Photostability
Researchers have developed a new red fluorescent calcium indicator called PinkyCaMP, designed to track neural activity while avoiding false signals caused by blue-light-induced photoswitching. This innovation, published in Nature Methods, utilizes the bright protein mScarlet to improve upon existing red calcium sensors, which have traditionally been dimmer and less photostable than their green counterparts. PinkyCaMP offers deeper tissue penetration and reduced phototoxicity, making it compatible with optogenetics and green fluorescent tools. The study demonstrated PinkyCaMP's superior brightness and photostability compared to other red sensors, with no false signals from blue light exposure. It was tested in human embryonic kidney cells and cultured mouse neurons, showing reliable calcium response tracking and compatibility with light-activated ion channels.
