GSTP1 Identified as Key Aging-Associated Biomarker in Prostate Cancer Progression
An integrative analysis has identified Glutathione S-transferase pi (GSTP1) as a significant aging-associated gene and a promising diagnostic candidate in prostate cancer (PCa). Researchers utilized a multi-omics approach, integrating transcriptome profiles, age-related gene screening, and machine learning techniques across four GEO datasets to pinpoint key diagnostic biomarkers. The study found that aging-related genes are highly enriched in pathways relevant to PCa, with GSTP1 demonstrating exceptional diagnostic performance. Unlike its overexpression in many other cancers, PCa is characterized by a frequent and early loss of GSTP1 expression, primarily due to hypermethylation of its gene promoter region. Experimental validation in PCa cell lines (PC3 and DU145) showed that depleting GSTP1 promotes oncogenic phenotypes, including increased proliferation, migration, and invasion, while attenuating senescence-associated markers. This suggests GSTP1 plays a crucial role in maintaining genomic integrity and ...
