Moderna and Merck Announce Positive Phase 3 Trial Results for Personalized mRNA Cancer Vaccine in Melanoma
Moderna and Merck have announced positive results from a Phase 3 clinical trial for intismeran autogene, an individualized mRNA-based cancer vaccine, when combined with the immune checkpoint inhibitor pembrolizumab (Keytruda®). The trial focused on patients with high-risk melanoma whose tumors had been completely removed by surgery. The combination therapy significantly extended recurrence-free survival and distant metastasis-free survival compared to Keytruda alone. This marks the first positive Phase 3 trial for an individualized neoantigen therapy and an mRNA-based cancer therapy. The vaccine works by analyzing a patient's tumor to identify unique genetic mutations, called neoantigens, which are then encoded into a personalized mRNA vaccine. This vaccine instructs the immune system to target these specific cancer markers, while Keytruda helps sustain the immune response by blocking the PD-1 checkpoint.
