What's Happening?
Avenzo Therapeutics, a clinical-stage biotechnology company, announced that it will present preliminary Phase 1 results from its ORION-1 study of AVZO-023 at the 2026 San Antonio Breast Cancer Symposium (SABCS). AVZO-023 is a selective cyclin-dependent
kinase 4 (CDK4) inhibitor being developed for hormone receptor-positive, human epidermal growth factor receptor 2-negative (HR+/HER2-) advanced or metastatic breast cancer. The presentation will detail preliminary safety, tolerability, and efficacy data for AVZO-023 when used in combination with fulvestrant. AVZO-023 is designed to inhibit CDK4/cyclin D-mediated signaling, a key driver of tumor cell proliferation, while sparing CDK6 to potentially reduce hematologic toxicity and improve tolerability in combination with other therapies.
Why It's Important?
HR+/HER2- metastatic breast cancer remains a significant challenge, and new treatment options are crucial for patients who may develop resistance to existing therapies. AVZO-023's selective inhibition of CDK4, while sparing CDK6, could offer a differentiated safety and efficacy profile compared to current CDK4/6 inhibitors. Improved tolerability could allow for more effective combination regimens, potentially extending the duration of treatment and improving patient outcomes. The ORION-1 study also explores a triplet combination with AVZO-021 (a selective CDK2 inhibitor) and fulvestrant, aiming to target both primary tumor proliferation and key resistance mechanisms. This multi-pronged approach could address unmet needs in patients who have progressed on other treatments.
What's Next?
The preliminary Phase 1 results will be presented at the 2026 San Antonio Breast Cancer Symposium in December. These findings will provide initial insights into the safety and potential efficacy of AVZO-023. Following the Phase 1 portion, the ORION-1 study will continue into Phase 2, further evaluating AVZO-023 in combination with endocrine therapy and potentially with AVZO-021. The data from these studies will be critical for guiding the further development and potential regulatory pathway for AVZO-023. Avenzo Therapeutics holds exclusive global rights, excluding Greater China, for AVZO-023 from Allorion Therapeutics, indicating a strategic focus on major international markets.
Beyond the Headlines
The development of highly selective CDK inhibitors like AVZO-023 reflects a growing trend in oncology towards precision therapies that minimize off-target effects. By specifically targeting CDK4 and sparing CDK6, Avenzo aims to reduce common side effects such as neutropenia, which can limit the use of existing CDK4/6 inhibitors. This approach could lead to better patient compliance and improved quality of life during treatment. The exploration of triplet combinations, including a CDK2 inhibitor, also highlights the evolving understanding of resistance mechanisms in breast cancer and the need for multi-target strategies to overcome them. This research could pave the way for more personalized and effective treatment regimens for patients with advanced breast cancer.
