Arrowhead Pharmaceuticals, Inc. announced interim topline data from its Phase 1/2a clinical trial for ARO-DIMER-PA, an investigational RNA interference (RNAi) therapeutic. This drug is designed as a potential treatment for atherosclerotic cardiovascular
disease (ASCVD) caused by mixed hyperlipidemia. ARO-DIMER-PA is unique as it is the company's first dual-functional RNAi molecule, engineered to simultaneously silence the expression of two genes: proprotein convertase subtilisin kexin 9 (PCSK9) and apolipoprotein C3 (APOC3). In participants with mixed hyperlipidemia, single doses of ARO-DIMER-PA achieved significant mean maximal reductions: 72% in serum PCSK9 and 88% in APOC3. This dual gene silencing led to favorable changes in lipids and lipoproteins, including a 54% reduction in LDL-C, 73% in triglycerides (TG), 61% in non-high-density lipoprotein (non-HDL) cholesterol, and 50% in Apolipoprotein B (ApoB). The study reported injection site events and headaches as the most common treatment-emergent adverse events, with no drug-related serious adverse events.
