The University of Colorado Anschutz Medical Campus is developing a functional precision medicine platform aimed at personalizing treatments for rare cancers. Led by Alice Soragni, PhD, a professor of Biomedical Informatics and Neurosurgery and the Marsico
Chair in Excellence in Functional Precision Medicine, the initiative focuses on creating patient-derived tumor organoids. These miniature 'tumor avatars,' grown from a patient's own tumor cells, are used to test responses to various drugs. The platform, detailed in Nature Protocols, utilizes label-free imaging and artificial intelligence to continuously track individual organoid responses, identifying differences that conventional methods might miss. The goal is to integrate this organoid testing into clinical care, moving it from a research tool to a validated clinical assay that can inform treatment decisions for patients with hard-to-treat tumors. This approach is particularly crucial for rare cancers, where standard treatments are often non-existent due to a lack of sufficient data.

Once validated, the results from these assays can be reported clinically, allowing physicians to use the findings to support treatment decisions. The initiative is starting with a streamlined version of a one-week-turnaround mini-ring organoid assay. In the long term, the data derived from this screening platform, combined with DNA and RNA sequencing and other molecular characterizations, is expected to help identify new biomarkers for drug responses, potentially leading to simpler tests and better medicines for a broader patient population.
