What's Happening?
Evecxia Therapeutics, a clinical-stage neuroscience company, has announced positive data from its Phase 1b PK/PD trial for brain serotonin target engagement. The trial focused on depression patients already undergoing selective serotonin reuptake inhibitor/serotonin-norepinephrine
reuptake inhibitor (SSRI/SNRI) therapy. The study demonstrated that EVX-301, an intravenous infusion of 5-HTP, successfully entered the brain, elevated biomarkers of serotonin synthesis, and increased serotonin activity beyond the effects of existing SSRI/SNRI treatments. These findings corroborate the target 5-HTP plasma exposure of approximately 100 ng/mL, which is considered the putative therapeutic target. The adjunctive EVX-301 treatment was generally well-tolerated, with most adverse events being mild or moderate and consistent with increased brain serotonin activity. The trial was conducted at PARC Clinical Research in Adelaide, Australia.

These positive Phase 1b results are significant for the development of new treatments for psychiatric conditions, particularly depression and obsessive-compulsive disorder (OCD). What's Next?
Following these positive Phase 1b results, Evecxia Therapeutics plans to initiate Phase 2 development for its lead candidate, EVX-101, in the first half of 2027. EVX-101 is an oral, gastro-retentive, sustained-release tablet formulation of 5-HTP and low-dose carbidopa, designed to be an adjunctive therapy for OCD patients who respond inadequately to SSRI treatment. The company believes that EVX-101, backed by extensive clinical data, addresses multiple unmet needs in psychiatry and can seamlessly integrate into existing psychiatric care and reimbursement infrastructures globally. The upcoming Phase 2 trials will further evaluate the efficacy and safety of EVX-101, moving it closer to potential market availability and offering a new therapeutic avenue for challenging psychiatric conditions.
