A recent study has introduced an innovative in-situ dual-lighting system (ISDLS) designed to improve the detection of HER2, a protein associated with certain types of cancer. The system combines molecular beacon-based loop-mediated isothermal amplification
(LAMP) and tyramide signal amplification (TSA)-based immunohistochemistry (IHC) to detect HER2 nucleic acid and protein in a single pathological section. This method aims to address the limitations of current diagnostic techniques, which often require confirmation through in situ hybridization (ISH) due to the limited reliability of IHC alone. The study demonstrated that ISDLS could achieve a 100% detection rate in clinical IHC ≥ 1+ and fluorescence in situ hybridization (FISH) positive sections, and a 77.8% detection rate in FISH negative sections, all within a single day.
