Researchers from Weill Cornell Medicine and Wake Forest University School of Medicine have discovered a potential method to prevent chemotherapy-induced peripheral neuropathy (CIPN), a common side effect of cancer treatment. The study, published in Science
Translational Medicine, reveals that chemotherapy activates a stress-sensing system in immune cells, leading to inflammation and nerve injury. By targeting the IRE1α-XBP1 pathway, which is responsible for this stress response, researchers found that they could reduce nerve damage in mice. A drug that blocks IRE1α, currently in phase 1 clinical trials, showed promising results in mitigating pain and preserving nerve health in mice treated with chemotherapy.

Chemotherapy-induced peripheral neuropathy affects up to half of all chemotherapy patients, causing significant pain and potentially leading to reduced or discontinued treatment.
